چکیده

By WENDELL F. RossE, THOMAS WALDMANN AND ELEANOR Huu. H YPOXIA AND ANEMIA have been known for many years to stimulate erythropoiesis in mammals, including man.1 In recent years, these stimuli have been shown to have their effect through a humoral factor, erythropoietin.2 J acobson, et al. have suggested that lack of oxygen, relative to the metabolic needs of the animal, is the fundamental stimulus for erythropoiesis and that this relative lack is sensed by some portion of the body, resulting in the production of erythropoietin.3 The lack of oxygen in the sensitive tissues may be due to hypoxic environment, to reduction of circulating hemoglobin, to the histotoxic effects of chemicals or, on the other hand, to an increase in tissue oxygen needs due to elevation of the rate of metabolism. There is some evidence that this hypothesis may not hold for lower animals. Chronic hypoxia does not induce polycythemia in turtles,4 frogs5’6 or lizards,6 although this response is said to occur in newts7 and fish.8 No reticulocytosis is seen in response to acute or chronic anoxia in the turtle although evidence of increased erythropoiesis is seen in response to bleeding.4 The present experiments were designed to test the erythropoietic response of frogs to bleeding, hypoxia, and to cobaltous ion administration. The response was measured by determining the incorporation of thymidine-2-C” into circulating erythrocytes. This compound is incorporated into desoxyribonucleic acid only of dividing cells; is not exchanged, once incorporated, until the cell dies; and is not incorporated into the non-dividing cell.#{176} This same system was used to test the erythropoietic effect of various sera.

Effect of temperature on erythropoiesis and red cell survival in the frog.

Cum, h/I. J., AND T, A. WALDMANN. Effect uf temperature on er~~hropaiesis and Ted cell survival in the frog. Am. J. Physiol. 203 (3) : 401-403. I g62.-The effect of environmental temperature on erythropoiesis and erythrocyte life span was studied in frogs maintained at 4 C or 24-26 C. The red cell incorporation of radioiron was used as an index of the rate of erythropoiesis and diisopropylfluor...

p-ERK1/2 is a predictive factor of response to erythropoiesis-stimulating agents in low/int-1 myelodysplastic syndromes.

Serum erythropoietin level less than 100U/L and a transfusion requirement of less than 2 units per month are the best predictive factors for response to treatment by erythropoiesis-stimulating agents in low/int-1 myelodysplastic syndromes. To investigate the factors influencing the response to erythropoiesis-stimulating agents, we enrolled 127 low/int-1 myelodysplastic syndrome patients at diag...

Ferritin Elevation and Improved Responsiveness to Erythropoiesis-Stimulating Agents in Patients on Ferric Citrate Hydrate

Introduction In hemodialysis patients on ferric citrate hydrate, the increase in ferritin level is mainly due to the administration of the compound. We investigated possible other factors associated with ferritin level and how erythropoietin resistance index and erythropoiesis in those patients were affected. We looked at ferritin-elevating factors using data from a Japanese phase III long-term...

Mutual inhibition of murine erythropoiesis and granulopoiesis during combined erythropoietin, granulocyte colony-stimulating factor, and stem cell factor administration: in vivo interactions and dose-response surfaces.

We investigated the in vivo effects of erythropoietin (EPO) on granulopoiesis and, conversely, the effect of granulocyte colony-stimulating factor (G-CSF) treatment on erythropoiesis. Recombinant human EPO at four different doses in combination with recombinant human G-CSF also at four different doses was simultaneously administered for 7 days to splenectomized mice. In total, 16 different comb...
